Contactless Fault Indicator for MV Lines

The theoretical findings about methods of earth faults indication are elaborated in this thesis into form, which is applicable to design contactless indicator of failure states on MV lines. This design contains electromagnetic field sensors, evaluation device and software support. The higher effectiveness of clearing of fault and better health and asset protection is supposed by using of these indicators.

Upgrade of testing workplace for solar cells

The master‘s thesis is focused on the modification of measurement workplace for solar cells. In the first part of the thesis are introduced general problems of the solar energy and possible use in the international power supply, the details about the photovoltaic effect, processing of solar cells and their subsequent parameters. More further in the text the autor is concerned with causes of solar cells defects formation and representation of the most important defects. For defects detection are known the detection methods of solar cells, which are generally described in the text. The second part of the thesis includes detailed description of the LBIC method and workplace both the VUT Brno, and in Solartec, Ltd. The most important point of this part is project and description of innovated workplace from hardware and software realization. There is out of print the function and the description of the user interface and subsequently there are mentioned results gained from original and innovated workplace. In the conclusion are summarized whole activities of the master’s thesis, described and evaluated achieved results and outlined the direction, where would the other development of the LBIC workplace on VUT Brno be proceeding.
Molecular detection of cercarial dermatitis causative agents

Human cercarial dermatitis (CD), also called Swimmer's itch appears as a skin rash caused by skin penetration by larval stages of avian schistosomes. In the Czech Republic, the most common causative agens is genus Trichobilharzia. Since 2021, monitoring of CD is a part of a mandatory testing of bathing water quality. The official method used for CD detection is collecting and examination of aquatic snails under a light source. However, this method has many disadvantages, such as time demands, low sensitivity and difficult parasite determination. These disadvantages can be overcome by usage of alternative approaches, such as molecular methods based on environmental DNA (eDNA) filtration and isothermal amplification. Hence, the main goal of this thesis was development of molecular method based on eDNA filtration and cLAMP isothermal amplification for detection of CD causative agent in water. Based on molecular analysis a conserved region of target DNA was selected to create a cLAMP assay specific for genus Trichobilharzia. The cLAMP assay was confirmed to specifically amplify the DNA of at least four species of the genus Trichobilharzia. Internal audit of banks and its function during detection of fraudulent actions

This bachelor thesis is focused on the analysis of internal audit in bank and his function during the detection of fraud schemes. Firstly, there is a summary of audit evolution in the world and Czech republic then the thesis defines general concept of audit and its parts such as internal and external audits and their relation. Within internal audit there is explained its principle, characteristic of internal auditor and integration into the organizational structure. The process of internal audit in bank is important part of the thesis and all phases and work activities are explained. There are defined types of frauds and their perpetrators in the segment of fraudulent activities. The thesis highlights the position of internal audit amongst several kinds of detection methods. This final finding is supported by two real cases that are analyzed from the perspective of internal audit.
Diagnotics of the important apple viruses

Apple mosaic virus (ApMV), Apple stem grooving virus (ASGV), Apple stem pitting virus (ASPV) and Apple chlorotic leaf spot virus (ACLSV) are economically the most important viruses in pome fruit trees, which are distributed worldwide and can caused significantly yield reduction. The major control strategies (namely pathogen detection, exclusion by crop certification or quarantine, control in infected orchards by eradication from infected cultivars and rootstocks, etc.) rely heavily on accurate and sensitive detection methods and on perfect knowledge of pathogens. In the doctoral thesis the diagnostic method quantitative RT-PCR (qRT-PCR) was optimized for detection and quantification of four studied viruses. The results suggested that qRT-PCR method was the most reliable technique in comparison with conventional diagnostic methods DAS/I-ELISA and RT-PCR. In our study the concentration of ASGV, ASPV and ACLSV, measured by qRT-PCR, were stable during vegetation and in different plant tissue. Only the concentration of ApMV changed during vegetation in leaves and inner bark. This result indicates that changes of virus concentration observed by DAS/I-ELISA and RT-PCR in plant tissues are caused by other way (inhibitors, plant senescence, lower sensitivity, ect.) than by changes of virus concentrations in plant. Under the monitoring (at all 351 trees were tested) it was showed, that studied viruses were more spread in orchards and gardens then in wild apple trees. Selected virus isolates from wild apple trees and apples from orchards and gardens were sequenced and molecular variability was studied also with already published isolates. However individual isolates of studied viruses were similar. The variability associated with geographic origin or with type of planting has not been confirmed.
